A black Citroen and white Ford Transit van were involved in a collision just before 4pm on the A1075 at Ovington, between Dereham and Watton.
The East Anglian Air Ambulance took a woman with a serious head injury to the Norfolk and Norwich University Hospital.
An unconscious man with injuries to his abdomen and knee and a person suffering with spinal injuries were also taken to the hospital. A fourth casualty was taken to the Queen Elizabeth Hospital in King's Lynn.
The accident closed the road both ways between Church Road and Long Road.
